Resumo: Para melhorar a eficácia de fármacos como nifurtimox, benznidazol e ravuconazol pode-se utilizar sistemas autonanoemulsionáveis para entrega de fármacos (SNEDDS). O estudo teve como objetivos: (i) avaliar a toxicidade dos SNEDDS e dos respectivos excipientes utilizados no preparo dos SNEDDS Nfx 1 e 2, SNEDDS Bz 3 e SNEDDS Rv 4 em células H9c2, uma linhagem de cardiomiócitos de ratos neonatos, e Hep G2, uma linhagem de hepatócitos de carcinoma humano; e (ii) avaliar a atividade anti-Trypanosoma cruzi dos SNEDDS Nfx 2 e SNEDDS Rv 4 in vitro, utilizando como modelo experimental células H9c2 infectadas pelas cepas Y e Colombiana do T. cruzi. Os excipientes usados no preparo dos SNEDDS não induziram diminuição da viabilidade das células H9c2 e Hep G2 nas mesmas concentrações usadas nas formulações nas doses efetivas de tratamento (5 e 10 μM, para nifurtimox; e 2nM, para ravuconazol). No entanto, o Tween 80 usado no preparo das formulações SNEDDS Nfx 1 e SNEDDS Bz 3, mostrou-se tóxico nos testes de citotoxicidade em células Hep G2, nas concentrações entre 0,065% e 0,26% v/v, reduzindo a viabilidade em mais de 40%. Os SNEDDS Nfx 1 e SNEDDS Bz 3 foram tóxicos a partir da concentração de 25 μM referente ao fármaco livre, por isso não foram realizados os testes de atividade com estes SNEDDS. O SNEDDS Nfx 2 apresentou menor toxicidade em relação ao SNEDDS Nfx 1, apresentando toxicidade a partir da concentração de 50 μM; e o SNEDDS Rv 4 não apresentou toxicidade significativa. Com isso, os SNEDDS Nfx 2 e SNEDDS Rv 4 foram utilizados nos testes de atividade in vitro e apresentaram efeito inibitório superior aos respectivos fármacos livres sobre formas amastigotas das cepas Y e Colombiana do T. cruzi. Este estudo in vitro evidenciou que os SNEDDS podem ser formulações seguras e potencialmente mais ativas contra as formas amastigotas do parasito.
Resumo em outra língua: To improve the efficacy of drugs such as nifurtimox, benznidazole and ravuconazole can be used self-nanoemulsifying drug delivery systems (SNEDDS). The study aimed to: (i) to evaluate the toxicity of SNEDDS and their excipients used in the preparation of SNEDDS Nfx 1 and 2, SNEDDS Bz 3 and SNEDDS Rv 4 in H9c2 cells, a cardiomyocyte cell line of mice, and Hep G2, a line of human carcinoma hepatocytes; and (ii) to evaluate the anti-Trypanosoma cruzi activity of SNEDDS Nfx 2 and SNEDDS Rv 4 in vitro, using as an experimental model H9c2 cells infected by strains Y and Colombian T. cruzi. The excipients used in the preparation of SNEDDS not induced decrease in viability of Hep G2 cells and H9c2 the same concentrations used in the formulations in effective treatment doses (5 and 10 μM for nifurtimox, and 2nM for ravuconazole). However, Tween 80 used in preparation of the formulations SNEDDS Nfx 1 and SNEDDS Bz 3, showed toxic in cytotoxicity assays in Hep G2 cells at concentrations between 0.065% and 0.26% v / v, reducing the viability of more than 40%. The SNEDDS Nfx 1 and SNEDDS Bz 3 were toxic at concentrations of 25 μM refering to the free drug, because of that the activity tests with these SNEDDS were not realized. The SNEDDS Nfx 2 showed lower toxicity compared to SNEDDS Nfx 1, with toxicity from the concentration of 50 uM; and SNEDDS Rv 4 showed no significant toxicity. Thus, the SNEDDS Nfx 2 and SNEDDS Rv 4 were used in vitro activity tests and showed inhibitory effect higher than the respective free drugs on amastigote forms of strains Y and Colombian T. cruzi. This in vitro study showed that the SNEDDS can be safe and potentially more active formulations against the amastigote forms of the parasite.
