dc.description.abstractNeutrófilos prevalecem nas primeiras horas após a infecção por Leishmania e representam, portanto, a primeira população celular a migrar para o local do inóculo, sendo cerca de 80 a 90% do infiltrado celular e, dessa forma, pode contribuir para resposta imune protetora na infecção por este parasito. Entretanto, diferentes estudos mostraram que neutrófilos podem auxiliar no estabelecimento da infecção, via inativação dos mecanismos leishmanicidas de macrófagos. Considerando que a maior parte dos trabalhos aborda a infecção por Leishmania major, torna-se de grande relevância o estudo do papel de neutrófilos na infecção por outras espécies de Leishmania. Neste sentido foi avaliada, in vitro, a resposta imune de neutrófilos na presença de diferentes espécies de Leishmania. Para isso, neutrófilos obtidos de medula óssea de camundongos C57BL/6 e isolados por gradiente de Percoll® foram cultivados na presença de Leishmania amazonensis, Leishmania braziliensis ou L. major, por 3 horas a 37°C / 5%CO2. Primeiramente, foi avaliada a viabilidade dos neutrófilos infectados por parasitos do gênero Leishmania, onde os resultados mostraram que o percentual de células viáveis não foi alterado, independentemente da espécie de Leishmania. Em seguida, foi avaliada a capacidade fagocítica através da marcação dos parasitos com éster de succinimidil-carboxifluoresceína (CFSE) e os resultados mostraram maior percentual de fagocitose de L. braziliensis por neutrófilos quando comparados a L. amazonensis ou L. major. Posteriormente, foi avaliado o perfil funcional dessas células através da avaliação da expressão de óxido nítrico (NO) e espécies reativas de oxigênio (EROs) intracelulares, por citometria de fluxo, e os resultados demonstraram maior expressão tanto de NO como de EROs por neutrófilos infectados por L. braziliensis quando comparados a L. amazonensis ou L. major. Adicionalmente, foi avaliado a expressão de moléculas de ativação (CD11b/CD49d/CD62L e TLR2/TLR4/TLR9) e os resultados mostraram diminuição da expressão de CD62L e aumento na expressão de TLR2 e TLR9 em neutrófilos infectados pelas três espécies de Leishmania. Além disso, foi observado aumento na expressão de TLR4 em neutrófilos infectados por L. braziliensis, quando comparado à população sem estimulo, bem como, aumento de TLR2 quando comparado com as demais espécies. Posteriormente foi realizada a dosagem dos níveis de quimiocinas (MCP-1, MCP-3, MIP-1α, MIP-1β e RANTES) no sobrenadante de cultura e os resultados mostraram que apenas neutrófilos infectados por L. amazonensis ou L. braziliensis produziram MIP-1α, além disso, independente do estímulo, neutrófilos foram capazes de produzir MIP-1β. Por fim, os resultados da avaliação do perfil de expressão intracelular das citocinas IL-12, TNF-α, IL-10 e TGF-β mostraram maior expressão de TNF-α por neutrófilos infectados pelas três espécies do parasito. Além disso, esse mesmo resultado foi observado em neutrófilos expressando IL-10 infectados por L. amazonensis ou L. braziliensis, bem como, em neutrófilos expressando IL-12p40 infectados por L. amazonensis, quando comparados aos neutrófilos sem estímulo. Nenhuma diferença foi observada para a citocina TGF-β nos neutrófilos infectados pelas três espécies do parasito. Em conjunto, os dados demonstraram que neutrófilos podem ter papel importante para o controle da infecção, bem como no estabelecimento e direcionamento de resposta protetora. Além disso, pode-se sugerir que L. braziliensis foi mais eficiente na indução de perfil de ativação e expressão de mediadores inflamatórios quando comparado às demais espécies, o que indica um possível mecanismo de controle da infecção desencadeado por neutrófilos.pt_BR

dc.description.abstractenNeutrophils prevail during the first hours after infection by Leishmania and therefore represent one of the first cell populations to migrate to the site of the infection, representing about 80 to 90% of the cellular infiltrate and, thus can contribute to a protective immune response on infection by this parasite. However, different studies have shown that neutrophils may assist in establishment of infection, via inactivation of macrophage antileishmanial mechanisms. Whereas most studies address the infection by Leishmania major, it is of great importance to study the role of neutrophils in other species of Leishmania. In our study, we evaluated in vitro response of neutrophils in the presence of different Leishmania species. For this, neutrophils harvested from bone marrow of C57BL/6 mice and then purified by Percoll gradient were cultured in the presence of L. amazonensis, L. braziliensis or L. major for 3 hours at 37°C / 5%CO2. Initially, we evaluated the viability of neutrophils infected by parasites of the genus Leishmania. The results showed that the percentage of viable cells did not change, regardless of the species of Leishmania used in the infection. We, then, evaluated the phagocytic ability by stainning parasites with carboxyfluorescein succinimidyl ester (CFSE) and the results showed a higher percentage of neutrophil phagocytosis in the presence of L. braziliensis when compared to L. amazonensis or L. major. Subsequently, we assessed the functional activation of these cells through the evaluation of nitric oxide (NO) and reactive oxygen species (ROS) intracellular expression by flow cytometry. Our results showed an increased expression of both NO and ROS by neutrophil infected with L. braziliensis when compared to L. amazonensis or L. major. In addition, we evaluated the expression of activation molecules (CD11b/CD49d/CD62L and TLR2/TLR4/TLR9) and the results showed a decreased expression of CD62L and an increase in the expression of TLR2 and TLR9 on neutrophils infected by three Leishmania species. Moreover, we observed an increase in expression of TLR4 in neutrophils infected by L. braziliensis, when compared to the population without stimulation, as well as increased TLR2 when compared with the other species. In addition, we evaluated the levels of chemokines (MCP-1, MCP-3, MIP-1α, MIP-1β and RANTES) in the culture supernatant and the results showed that only neutrophils infected by L. amazonensis or L. braziliensis produced MIP-1α. Moreover, independently of the stimulus, neutrophils were able to produce MIP-1β. Finally, the results of IL-12, TNF-α, IL-10 and TGF-β intracellular expression showed increased levels of TNF-α in neutrophils infected by the three species of the parasite. Additionally, this same result was observed in neutrophils expression of IL-10 infected by L. amazonensis or L. braziliensis and, in neutrophils expression IL-12p40 infected by L. amazonensis, when compared to non stimulated neutrophils. No difference was observed for the cytokine TGF-β in neutrophils infected by the three species of the parasite. Together, these data demonstrated that neutrophils may have an important role in the control of infection, as well as establishing and directing a protective response. Furthermore, it can be suggested that L. braziliensis was more effective in inducing a profile activation and expression of inflammatory mediators when compared to other species, suggesting a possible mechanism of control neutrophils triggered by the infection.-
